ONE HEALTH APPROACH TOWARDS ELIMINATION OF RABIES - A 5-DAY TRAINING FOR VETERINARY PROFESSIONALS” (13th -17th SEPTEMBER, 2018)
Rabies always proves to be fatal and leads to one of the most torturous forms of death from any infectious disease. The problem of dog bites and, consequently, of rabies, has been an extremely contentious and neglected issue in Pakistan. Over 20,000 new cases of dog bite victims are treated annually in Pakistan, with over 2,000-5,000 rabies deaths annually. It especially affects children (below the age of 15 years) as they play in streets and are unable to run away or defend themselves against an attacking animal. Some of the bites are severe and disfiguring, as well as dangerous, if the victim is bitten on the head, face or neck. Occasionally, several rogue dogs maul small, defenseless children. Many families have been devastated by the loss of their child or a working family member. Knowledge, application and practices against rabies prevention are extremely poor among both physicians and general public. The University of Veterinary and Animal Sciences (UVAS) under its project entitled “In-service Training Facility of Advance Veterinary Education and Professional Development for Veterinary Professionals (IVPD)” in collaboration with Indus Hospital Karachi on 13th -17th September 2018 arranged five-day training workshop on “One Health Approach towards Control of Rabies”.
The participants of this training were of 5th, 7th and 9th semester students of Doctor of Veterinary Medicine (D.V.M) from main campus and Ravi Campus of UVAS, Lahore. The aim and overreaching objectives of the training was to learn and share the Indus Hospital Rabies control team’s experiences and to provide hands on training opportunity to D.V.M students for their capacity building to run an impactful Rabies Free Lahore campaign to share our hands in fulfilling WHO’s Vision 2030 of Rabies control.

FOREIGN CLINICAL TRAINING
BASIC CLINICAL AND DIAGNOSTIC SKILLS IN SMALL ANIMAL PRACTICE
by Dr. AAMIR ISLAM FROM USA
(04th – 08th June, 2018)
Hands On Training In Bovine Embryology
by Prof. Dr Kehuan Lu from China
(24th – 28th June, 2018)
One-week international hands on workshop on bovine embryology by the Department of Theriogenology in collaboration with IVPD project has been started in UVAS, Ravi Campus Pattoki under the supervision of foreign expert Prof. Dr. Kehuan Lu (Guangxi University, Nanning, China). The main objective of this workshop to equip the faculty with in vitro embryo production technology including follicle aspiration, oocytes grading, in-vitro maturation, in-vitro fertilization, Semen preparation, in-vitro culturing and grading of embryos. There are 15 participants from City Campus, Jhang Campus and Narowal Campus as well.
Objectives
The basic objective was hands on training of:
Different medias preparation for IVEP program
Ovaries collection and COCs aspiration
Searching and grading of COCs
Invitro maturation of COCs
Invitro fertilization COCs
Invitro sperm capacitation
Invitro culturing of presumptive embryos
Biosafety measure and QC of the embryology work station
Advances in bovine embryology and establishment of new techniques
SUGGESTIONS AND GUIDELINES BY PROF. KEHUAN LU
Ovaries should reach in the lab within 3-4 hours at 33-37oC temperature
Use 18 G needle and 10 ml syringes for aspiration of follicles
Only aspirate the follicles having size between 4-10 mm in diameter
For selection of good quality oocytes evaluate the no. of COCs layers and ooplasm homogeneity and color
For washing of oocytes use excessive media about 3-4 ml per drop in 35 mm dish (Don’t prepare drops for washing purpose)
Before each step preheat the media or prepare the drops and preheat the dishes for minimum 1 hour (2 hours are preferred)
From aspiration to placement in IVM media, all the steps should be completed within an hour
IVM timing should be 22 hours
In buffalo maturation stage is more critical. So use IVF-Bioscience media for IVM of Buffalo oocytes
Sperm oocyte co culture should be of 12 hours
Use 10ml semen capacitation media and two straws of semen. Make final concentration 2 million sperms per ml in the final IVF media (calculate volume accordingly)
After 36 hours of IVC, examine the cleavage. Following stages could be observed a) 2-cell stage embryos b) 4-cell stage embryos c) 8 cell stage embryos. Only the embryos which are at >2 cell stage embryos are transferred to new IVC drop at this time to enhance blast rate. Place it in incubator for 7 days.
All bench work after IVM like washing in IVF drops and denuding must be completed within 30 minutes
Don’t open the incubator frequently during culturing conditions
50 µl drops (for 20 oocyte or embryo) is enough for maturation, fertilization and culture or use 2 µl/oocyte or embryo.
Lab temperature should not be below than 25oC during working because oocyte get shock at lower temperature
Humidity should not be >50% in the lab
Prepare the glass pipettes before use for each step, place in steel boxes and sterilize or autoclave them
Pasteur pipette can be autoclaved for re-using if the media is not stuck in them.
Clean the stereo microscope stage before use each time
After each media/ drops preparation clean the safety cabinet
Label the petri dishes at bottom instead of on their covers to avoid any confusion
Quality control must be applied in the lab
Make your own lab manual
Purchase Osmometer to check the osmolarity of media
You can store the media at 4-10oC for 1 month without adding hormones and serum.
Sterilize the incubator monthly. Can use ethanol (70%)
Prepare your own washing media and semen preparation media
Can use 1ml syringe for picking oocyte rather than mouth pipette
Can use filter in mouth pipette to avoid contamination
